This stage featured eight winners from the third preliminary round and eight top-ranked UPL teams from last season. Twelve elite representatives were joined by three First League teams and one from the Second League.

Among the lower-division teams, Bukovyna and Viktoriya advanced to the next stage. Bukovyna ended the surprise run of Probiy and will make its first appearance in the Ukrainian Cup quarterfinals. Viktoriya secured their spot by drawing level late in extra time against LNZ and winning in a penalty shootout. FC UKSA, on the other hand, fell to Oleksandriya.

Dynamo's away match in Poltava proved tough, with the Kyiv side scoring the winning goal only in extra time. Shakhtar also earned a narrow win over Zorya but did so within the regular 90 minutes.

Other matchups included Rukh’s victory in the Lviv derby, ensuring their debut in the quarterfinals, Polissya overcoming Kryvbas, and Veres securing a win over Obolon in extra time.
